Integrin Addressin Interaction

Integrin addressin interaction is the specific binding between leukocyte integrins and addressins, adhesion molecules that guide immune cells to particular vascular tissues. Chemokine signaling activates integrins, increasing their affinity and enabling firm attachment to endothelial addressins such as ICAM-1, VCAM-1, or MAdCAM-1 before cells migrate across the vessel wall. In pharmacology, this interaction is an important target for therapies that modify leukocyte trafficking and inflammation. Agents that block integrins or their addressins can reduce immune-cell recruitment in disorders such as multiple sclerosis and inflammatory bowel disease, while also illustrating how altered cell adhesion can produce therapeutic benefits and safety risks.

Integrins bind ligands and transmit information from outside the cell to inside or vice-versa through an "outside-in signaling" or "inside-out signaling." In "outside-in signaling," external factors in the extracellular space bind to exposed ligand binding sites on integrins. This causes the inactive protein to undergo a conformational change to become active. Integrins are often clustered on the cell membrane. Repetitive and regularly spaced ligand binding events provide an effective stimulus.

Static adhesion assay is a powerful tool that can be used to model the interactions between T lymphocytes and other cell types. Interactions are generated by injecting labeled T cells into wells coated with adhesion molecules, while a plate reader is used to quantify the number of adherent cells following serial washes.
