Krebs Solution Perfusion

Krebs solution perfusion is an ex vivo technique that circulates an oxygenated, physiologically balanced salt solution through an isolated organ or tissue to preserve its function for pharmacological study. The solution contains key ions and buffering components that help maintain osmotic balance, pH, and cellular excitability, while continuous perfusion delivers oxygen and nutrients and removes metabolic waste. Researchers use this preparation to measure contractile, electrical, vascular, or secretory responses to drugs under controlled conditions. It supports concentration-response experiments, assessment of receptor-mediated effects, and investigation of tissue pharmacodynamics without the complexity of whole-animal physiology.

In this video, we perform Langendorff retrograde perfusion of an isolated mouse heart. During the procedure, the cardiac activity of the heart is maintained by perfusing it retrogradely with a perfusion solution via the coronary arteries using a cannula inserted into the ascending aorta. The model is used to study the cardiac pathophysiology of the heart.
