Metformin

Metformin is a biguanide antidiabetic medication that lowers blood glucose and remains a first-line pharmacologic treatment for many people with type 2 diabetes. It primarily reduces hepatic gluconeogenesis, in part through effects on cellular energy metabolism and activation of AMP-activated protein kinase, while also improving peripheral insulin sensitivity and limiting intestinal glucose absorption. In pharmacology, metformin provides a model for studying how drug actions can reshape metabolic pathways without directly stimulating pancreatic insulin release. Its clinical use supports glycemic control and can reduce diabetes-related complications, while its pharmacokinetics, contraindications, and gastrointestinal adverse effects remain important considerations in treatment design.

Pancreatic cancer stem cells (CSCs) can be expanded in vitro using the anchorage-independent sphere culture technique, which represents a powerful tool to study CSC biology and can serve as the first step to develop novel CSC-targeting therapies. Here the methodology for expanding, analyzing and targeting of pancreatic CSCs is provided.

This protocol describes a technique for monitoring extracellular matrix rigidity changes by using particle-tracking microrheology (PTM). This is done by culturing 3D pancreatic tumor spheroids into ECM with embedded fluorescent probes for video particle-tracking and analysis.
