アシル化は、カルボニル炭素に結合した脱離基をアミノ基で置換します。
まず、求核性アミンがカルボニル炭素に接近して四面体中間体を形成します。その後、脱離基が失われると、カルボニル結合が再構築されます。
最後に、別のアミン分子がブロンステッド塩基として機能し、プロトン化アミドから水素を抜き取り、中和して遊離アミドにします。
したがって、アシル化はアミンの2つの当量を使用します:最初の等価物は求核剤として作用し、2番目の等価物は塩基として機能します。
