学習と記憶は、ショウジョウバエ(D.)からヒト1に保存された、環境への進化的に古代の適応メカニズムである。フルーツフライは、本質的な分子メカニズムを解剖するための強力な遺伝的ツールの広い範囲を提供しているので、学習と記憶の基本的な原則を研究するための堅牢なモデル生物です2。学習と記憶不可欠なrutabaga3健忘4dunce5遺伝子を同定した先駆的な遺伝子スクリーニング研究は、フルーツハエが食べ物、潜在的な仲間を見つけ、捕食者を避けるために鋭い嗅覚に依存しているので、嗅覚コンディショニングを利用しました6

学びと記憶のメカニズムを研究する嗅覚コンディショニングは、タリーとQuinn7,8による嗅覚T迷路の導入のおかげで、人気のパラダイムとなっています。続いて、視覚調節9、求愛コンディショニング10、回避光タクシー抑制アッセイ11、およびスズバチ露光調節12を含む様々なタイプの学習および記憶を測定する他の方法が提案されている。しかし、これらのアッセイのほとんどは、大学のワークショップでカスタム構築するか、ベンダーを通じて購入しなければならない複雑なセットアップを持っています。ここで説明するパラダイムは、いくつかの利用可能な供給で簡単に組み立てることができるハエの回避的な連想学習を研究するための単純な行動アッセイに基づいています。

1. 受動回避装置の準備

  1. 14 mL ポリプロピレン培養チューブの壁面に垂直な 4 mm の穴を開け、チューブ底部から 8 mm 離れたところにドリルします。
    メモ:最良の結果を得るには、電動ドリルと5/32ドリルビットを使用してください。
  2. 鋼製のユーティリティナイフを使用して、14 mLポリプロピレン培養チューブの上部を切断し、長さ45mmのチューブ底部を作成します。下の断片は、下部のコンパートメントとして機能します。
  3. 単一のエッジカミソリブレードを使用して1,000 μLの青いピペットチップの先端を切り取り、開口部を単一のフライの通過に十分な幅にします。青い先端の狭い部分を切り取って、12mmの断片を作成します。この断片を下の区画の4mm穴にしっかりと差し込みます。これはハエを移すための積み込みドックとして使用されます。
  4. 15mm透明なビニールチューブ5/8"ID( 材料表を参照)をカットしてカップリングを作成します。反対側の端からカップリングに上側と下側のコンパートメントを挿入し、下部コンパートメントを上部のコンパートメントにしっかりと取り付けます。
  5. 2本の調整可能なクランプを使用して、アセンブリを垂直スタンドに取り付けます。上部コンパートメントとしてショックチューブを使用してアセンブリを垂直方向に向けます。
  6. ショックチューブワイヤを電気刺激装置( 材料表参照)に接続して、感電を引き起こす。トレーニング期間は1分です。
    注:観察を容易にするために、ショックチューブの後ろに白い紙を置いて、装置の白い背景として機能させます。ショックコンパートメントの上に75 W相当のソフト電球を備えたランプを置きます。セットアップの前に、調節可能な腕の拡大鏡ランプを置きます。パッシブ回避装置の表現を 図1に示す。

2. 受動回避手順のためのハエの準備

  1. 3-4日齢 のD.メラノガスター または D.シミュラン は氷冷ブロックを使用して飛び、実験の24時間前に食品24時間(バイアルあたり1飛ぶ)を持つ個々のバイアルに移す前に、以前に説明した手順19に従う。
    注:ここで説明した実験は、3-4日齢の男性 対を比較しましたD.メラノガスターとD . シミュランで雌のハエ。
  2. 行動実験の前に、すべてのバイアルをコーディングします。このためには、各グループに文字を割り当てます(例えば、「A」、「B」、「C」など)、各フライは数字です。すべてのデータが記録され、分析された後にのみ、このコードを明らかにする。個々の変動に対抗するために、遺伝子型または治療ごとに少なくとも20ハエを使用してください。
    注: 実験と「ブラインド」解析を実行すると、フライとデータ分析のパフォーマンスを評価する際の偏りは除外されます。

3. 最初のトライアルを実施する

  1. 前に説明したフライマウス吸引器(材料表を参照)を使用して個々のバイアルからローディングドックを介して下側コンパートメントにハエをそっと移します。空気を吸って口のアスピレーターに1つのフライをそっと吸う。軽く装填ドックに吹き込んでフライを堆積します。
    注:キャッチとロード中に動物にストレスを与えないようにしてください。
  2. フライが下のコンパートメントにロードされた直後に、1分タイマーとストップウォッチを開始します。
    注: ストップウォッチは、遅延を測定し、ショックの数をカウントする集計カウンターに使用されます。
  3. ストップウォッチを押すと、フライがショックチューブに入ったときに、グリッド上にすべての足を配置して最初の待ち時間を記録します。刺激装置をオンにして、フライに感電を与えます。刺激パラメータは120ボルト、1000 msの持続時間、1パルス/秒(PPS)、トレーニング所要時間2000 msです。
  4. フライが衝撃管に再入りした場合、追加の衝撃を与えます。1分のトライアル中に、集計カウンターまたはArduinoベースのカウンターで受けた衝撃の数を記録 します(資料表を参照)。Arduinoベースのカウンターをご利用の場合は、以下の手順に従ってください。

  6. 70%エタノールで下側とショックコンパートメントを洗浄し、糸くずのない洗浄ティッシュ( 材料表を参照)で拭き取り、ヘアドライヤーで乾燥させます。次のフライでトライアルを繰り返します。
  7. 行動実験の後、水と無臭の洗剤で下のコンパートメントを清掃してください。下部コンパートメントとショックコンパートメントを70%エタノールで拭き取り、一晩で空気乾燥します。

4. 第2のトライアルを実施する

  1. 上記の手順を繰り返して第2の試用を行う(ステップ3)24時間後。前日と同じ順序でハエをテストします。

5. 結果の分析

  1. 動物の実験グループごとに、試験1と試験2の平均待ち時間、衝撃の平均回数、およびグルーミングバウトの持続時間を計算します。Tukeyのtest23を使用して、ポストホック分析との複数比較のために、2グループ比較またはANOVAのための学生t検定を実行します。

The assay worked equally well in D. melanogaster and D. simulans male and female flies, demonstrating that the paradigm could be adapted to different D. species. The changes in fly behavior characterized by increased latencies and decreased number of shocks were statistically significant in the second trial and would strengthen with subsequent trials. Interestingly, if naïve flies were habituated to the apparatus without electric shock, they would enter the upper compartment a little faster on the second and the third trials. However, the decrease in latencies was not statistically significant (data not shown). No statistically significant differences were observed between sexes, although female flies had somewhat longer latencies and received slightly more shocks. This difference could be due to a combination of factors, including females' failure to associate the shock with the upper compartment, a stronger geotaxis, or possibly because females are slightly larger and slower than males.
