Reflectin A1

Reflectin A1 is a cephalopod structural protein that contributes to dynamic iridescence, making it a model for studying how biomolecules control light at the nanoscale. As an intrinsically disordered protein, it can undergo phosphorylation and electrostatic interactions that regulate its condensation, assembly, and association with cellular membranes; the resulting nanostructures alter refractive-index contrast and the wavelengths of reflected light. Biochemical studies of Reflectin A1 clarify how protein sequence, charge, and organization produce tunable optical properties. This knowledge supports the development of bio-inspired photonic materials, adaptive coatings, optical sensors, and other technologies that mimic cephalopod camouflage.

Zebrafish retinal regeneration has mostly been studied using fixed retinas. However, dynamic processes such as interkinetic nuclear migration occur during the regenerative response and require live-cell imaging to investigate the underlying mechanisms. Here, the experimental protocols are described for preparing Drosophila at different developmental stages and performing longitudinal optical imaging of Drosophila heartbeats using a custom optical coherence microscopy (OCM) system. The cardiac morphological and dynamical changes can be quantitatively characterized by analyzing the heart structural and functional parameters from OCM images.
