Eae Gene

The eae gene is a bacterial virulence gene that encodes intimin, an outer-membrane adhesin used by enteropathogenic and enterohemorrhagic Escherichia coli to attach to intestinal epithelial cells. During infection, these bacteria inject the translocated intimin receptor, Tir, into host cells through a type III secretion system; intimin then binds Tir, producing intimate attachment, actin rearrangement, pedestal formation, and destruction of nearby microvilli. Detecting eae with polymerase chain reaction and other molecular assays helps identify attaching-and-effacing pathogens in clinical, food, and environmental samples. Studying this gene clarifies bacterial colonization mechanisms and supports surveillance of diarrheal disease risks.

Source: Schmitz, K., et al., Bioluminescence and Near-infrared Imaging of Optic Neuritis and Brain Inflammation in the EAE Model of Multiple Sclerosis in Mice. J. Vis. Exp. (2017).This video demonstrates the method for visualizing and monitoring Experimental Autoimmune Encephalomyelitis or EAE-induced inflammation in the optic nerve using bioluminescence imaging. It includes positioning the mouse, reagent injection, and capturing sequential images to monitor changes in bioluminescence over time.

The transplantation of neural stem/progenitor cells (NPCs) holds great promises in regenerative neurology. The systemic delivery of NPCs has turned into effective, low invasive, and therapeutically very efficacious protocol to deliver stem cells in the brain and spinal cord of rodents and nonhuman primates affected by experimental chronic inflammatory damage of the central nervous system.

Certain mouse strains are able to resist induction of experimental autoimmune encephalomyelitis (EAE) with myelin basic protein. Described here is a simple immunization protocol that reverses the unresponsiveness and induces paralytic disease in several typical EAE resistant mouse stains.

Determining Blood-Brain Barrier Disruption Using Fluorescent Dyes in an EAE Mouse Model

2025

This video demonstrates a procedure for assessing blood-brain barrier disruption in an EAE mouse model. Fluorescent dyes of different sizes are injected retro-orbitally and circulate through the brain's blood vessels. The smaller dye leaks through gaps into the brain tissue, while the larger dye enters only if the disruption is severe, correlating with the extent of barrier damage.
