Transparent Body

A transparent body is a biological condition in which an organism or tissue allows substantial light to pass through, revealing internal structures and reducing visual contrast. Transparency arises when pigments are sparse and tissues, fluids, and cellular components have similar refractive indices, limiting light scattering; specialized structures may further enhance this effect. In biology, transparent organisms and engineered specimens support direct observation of anatomy, development, physiology, and disease processes with minimal dissection. Studying body transparency also clarifies camouflage, optical adaptation, and how biological materials interact with light, providing useful models for microscopy and noninvasive imaging.

Here we present a fluorophore based imaging technique to detect cell viability on a non-transparent titanium scaffold as well as to detect glimpses of the scaffold impurities. This protocol troubleshoots the drawback of imaging cell-cell or cell-metal interactions on non-transparent scaffolds.

Non-fouling PEG silane monolayer was desorbed from individually addressable ITO electrodes on glass by application of a reductive potential. Electrochemical stripping of PEG-silane layer from ITO microelectrodes allowed for cell adhesion to take place in a spatially defined fashion, with cellular patterns corresponding closely to electrode patterns.
