Viral Envelope

A viral envelope is a lipid membrane surrounding some viruses, helping them enter host cells and evade or interact with the immune system. Typically acquired from a host-cell membrane during viral budding, the envelope contains viral glycoproteins that bind specific cell-surface receptors and can trigger membrane fusion, releasing the viral genome into the cell. Envelope composition and stability influence host range, transmission, environmental sensitivity, and susceptibility to detergents or disinfectants. Understanding these features supports the development of vaccines, antiviral drugs, diagnostic assays, and strategies to study viral entry, assembly, and spread.

Outer Layers of the Cell Envelope

2025

The outermost layers of prokaryotic cells play a critical role in their survival, virulence, and interaction with the environment. These layers, often composed of polysaccharides, polypeptides, or proteins, form protective and adhesive structures that vary in organization and function.Capsules and Slime LayersCapsules are highly organized, tightly bound layers that firmly attach to the bacterial cell wall. Capsules are usually made of polysaccharides, though some are made of polypeptides. These...

The protocol describes a high-throughput approach to determining structures of membrane proteins using cryo-electron tomography and 3D image processing. It covers the details of specimen preparation, data collection, data processing and interpretation, and concludes with the production of a representative target for the approach, the HIV-1 Envelope glycoprotein. These computational procedures are designed in a way that enables researchers and students to work remotely and contribute to data...

An In Vitro Recombinant Virus Reporter System for the Detection of Viral Infection

2025

This video demonstrates an in vitro technique to detect hepatitis B virus (HBV) infection via a recombinant HBV reporter system. Upon infecting host cells, the recombinant viral genome expresses the luciferase reporter enzyme, which produces bioluminescence upon oxidizing its substrate — helping detect viral infection.

An Assay to Evaluate the Effect of Test Compounds on Viral Entry and Fusion in Host Cells

2025

This video demonstrates an assay to evaluate antiviral agents that target specific steps of the viral entry into host cells and fusion with the endosome. The cells are incubated at a low temperature and then infected with HCV. The low temperature permits viral binding to host cells but inhibits internalization via endocytosis. The test antiviral compound is added, and the cells are incubated at physiological temperature. This shift to a higher temperature facilitates viral entry and allows...
