Cecal Injection

Cecal injection is a surgical technique that introduces tumor cells or other experimental materials into the cecum, creating a localized model for studying colorectal cancer in its native tissue environment. During the procedure, the cecum is exposed and a controlled injection places cells within or beneath the cecal wall, where they can establish an orthotopic tumor and interact with surrounding stromal, vascular, and immune components. This approach helps researchers investigate tumor growth, invasion, metastasis, and treatment response under conditions that more closely reflect the primary tumor site than ectopic models. It is useful for evaluating cancer mechanisms, therapeutic strategies, and disease progression in vivo.

Experimental sepsis can be induced in mice using the cecal ligation and puncture (CLP) method. Current protocols to assess autophagy in vivo in the context of CLP-induced sepsis are presented here: A protocol for measuring autophagy using (GFP)-LC3 mice, and a protocol for measuring autophagosome formation by electron microscopy.

Intracecal Tumor Cell Injection: A Technique to Deliver Cancer Cells for Establishing Orthotopic Colorectal Cancer Mouse Model

The video describes the technique for intracecal injection of cancerous cells to develop an orthotopic mouse model with highly uniform tumors. Therefore, the orthotopic mouse model allows realistic simulation of metastasis, providing a valuable tool to study the phenotype changes during tumor growth and dissemination.

This protocol presents the operative details of cecal ligation and puncture (CLP) in a mouse model of sepsis. CLP is one of the most widely used techniques to create an animal model of sepsis. Therefore, a standardized CLP protocol is required for the attainment of reliable research results.
