Spectral Compensation

Spectral compensation is a quantitative correction method that separates overlapping fluorescence signals so each measured channel more accurately represents its intended fluorophore. In multicolor flow cytometry, fluorophores emit across more than one detector, creating spillover; compensation uses single-stained controls to calculate and subtract the predictable contribution of one fluorophore from other channels. This process enables reliable identification and comparison of cell-surface and intracellular markers in complex samples. In cancer research, accurate compensation supports immunophenotyping of tumor and immune cells, characterization of heterogeneous populations, and evaluation of biomarkers or treatment responses, strengthening conclusions from multiparameter cytometry experiments.

This article describes spectral cytometry, a new approach in flow cytometry that uses the shapes of emission spectra to distinguish fluorochromes. An algorithm replaces compensations and can treat auto-fluorescence as an independent parameter. This new approach allows for the proper analysis of cells isolated from solid organs.

Photoacoustic ophthalmology (PAOM), an optical-absorption-based imaging modality, provides the complementary evaluation of the retina to the currently available ophthalmic imaging technologies. We report the using of PAOM integrated with spectral-domain optical coherence tomography (SD-OCT) for simultaneous multimodal retinal imaging in rats.
