Neutral Lipid Measurement

Neutral lipid measurement is the quantitative analysis of nonpolar lipids, including triglycerides and cholesteryl esters, in chemical, biological, or food samples. Depending on the method, these lipids are extracted into organic solvents, separated by chromatography, or detected with fluorescent dyes that partition into hydrophobic lipid phases and produce a signal related to lipid abundance. Accurate measurements help characterize lipid composition, energy storage, sample quality, and metabolic changes. In chemistry and related research, neutral lipid assays support studies of lipid metabolism, formulation development, food analysis, and the evaluation of cellular responses to environmental or pharmacological conditions.

This time-saving technique offers an alternative to traditional gravimetric-based lipid quantification protocols. In this video, we demonstrate in vitro staining of neutral lipids in hepatocytes to detect hepatic steatosis or excessive fat accumulation in liver. Excessive fat is accumulated in the cells in form of lipid droplets. Oil Red O is a lipid soluble dye that binds to intracellular neutral lipids in these lipid droplets. When observed under a light microscope, these intracellular lipid droplets appear intensely red in color.
