Pharmacokinetics Phase

The pharmacokinetics phase describes how a drug’s concentration changes in the body over time, providing the basis for understanding its exposure and duration of action. It follows the processes of absorption into the bloodstream, distribution among tissues, metabolism that chemically modifies the compound, and excretion, collectively known as ADME; these processes shape concentration-time profiles and measures such as clearance and half-life. In clinical research, pharmacokinetic analysis helps determine dosing intervals, assess bioavailability, compare formulations, and evaluate how patient characteristics or interactions affect drug exposure. This information supports safer, more effective dose selection and the interpretation of clinical outcomes.

A phase diagram combines plots of pressure versus temperature for the liquid-gas, solid-liquid, and solid-gas phase-transition equilibria of a substance. These diagrams indicate the physical states that exist under specific conditions of pressure and temperature and also provide the pressure dependence of the phase-transition temperatures (melting points, sublimation points, boiling points). Regions or areas labeled solid, liquid, and gas represent single phases, while lines or curves represent...

Whether solid, liquid, or gas, a substance's state depends on the order and arrangement of its particles (atoms, molecules, or ions). Particles in the solid pack closely together, generally in a pattern. The particles vibrate about their fixed positions but do not move or squeeze past their neighbors. In liquids, although the particles are closely spaced, they are randomly arranged. The position of the particles are not fixed—that is, they are free to move past their neighbors to occupy...

Pharmacokinetics is a scientific discipline that focuses on the journey of a drug within the body, encompassing four key stages: absorption, distribution, metabolism, and elimination. The first stage, absorption, involves the drug's transfer into the bloodstream. Several factors dictate the extent and speed of this process. For example, the liver often metabolizes oral drugs before they reach systemic circulation, leading to only partial absorption. In contrast, intravenous (IV) administration...

Pharmacokinetic Models: Overview

2024

Pharmacokinetic models utilize mathematical analysis to achieve a detailed quantitative understanding of a drug's life cycle within the body. They are instrumental in simulating a drug's pharmacokinetic parameters, predicting drug concentrations over time, optimizing dosage regimens, linking concentrations with pharmacologic activity, and estimating potential toxicity. There are three primary types of models: empirical, compartment, and physiological. Empirical models, with minimal assumptions,...
