Immunomagnetic Purification

Immunomagnetic purification is a cell-separation technique that isolates specific cells from a mixed population using antibody-coated magnetic particles, enabling researchers to obtain enriched or depleted cell fractions. The antibodies bind surface markers on target cells, and applying a magnetic field captures labeled cells while unlabeled cells remain in suspension; positive selection and negative selection therefore produce different sample compositions. In developmental biology, this approach helps separate progenitor, stem, or differentiated cell populations for lineage analysis, gene-expression studies, and functional assays. Its speed and specificity support studies of cell fate, tissue formation, and developmental heterogeneity.

In this video, we demonstrate the isolation of monocytes from whole human blood by immunomagnetic negative separation. The isolated monocytes can be used for further downstream analysis.

Negative Immunomagnetic Selection: A Method to Purify B-cells from Peripheral Blood Mononuclear Cells

2023

The video explains a way to purify B-cells by immunomagnetic targeting and separation from peripheral blood mononuclear cells suspension. The method involve labeling unwanted cell types with specific antibodies or ligands (linked to magnetic particles) targeting specific cell surface proteins, to be depleted from the heterogenous cell suspension by magnetic separation.

We present the techniques required to isolate the stromal vascular fraction (SVF) from mouse inguinal (subcutaneous) and perigonadal (visceral) adipose tissue depots to assess their gene expression and collagenolytic activity. This method includes the enrichment of Sca1high adipose-derived stem cells (ASCs) using immunomagnetic cell separation.

Plasmid Purification

2023

Plasmid purification is a technique used to isolate and purify plasmid DNA from genomic DNA, proteins, ribosomes, and the bacterial cell wall. A plasmid is a small, circular, double-stranded DNA that is used as a carrier of specific DNA molecules. When introduced into a host organism via transformation, a plasmid will be replicated, creating numerous copies of the DNA fragment under study. In this video, a step-by-step generalized procedure is described for how to perform plasmid purification.

Gel Purification

2023

Gel purification is used to recover DNA fragments after electrophoretic separation. DNA recovery from an agarose gel includes three basic steps: binding, washing and eluting from a silica column. DNA is believed to bind to silica in the presence of high salt via a salt bridge. Following binding, DNA is washed of impurities and eluted under low salt conditions disrupting this interaction. This video goes through a step-by-step, generalized procedure for cutting out a band from the gel, gel...
