Integrin Alpha Six Beta One

Integrin alpha six beta one is a cell-surface adhesion receptor that connects cells to laminin-rich extracellular matrices and helps organize tissue architecture during development. This heterodimeric transmembrane protein binds laminins through its extracellular domain, while its cytoplasmic tails associate with adaptor proteins and the actin cytoskeleton to transmit mechanical and biochemical signals into the cell. In developmental biology, integrin alpha six beta one supports cell polarity, migration, survival, and differentiation in tissues such as epithelia, nervous system structures, and developing organs. Studying its regulation clarifies how extracellular matrix interactions guide morphogenesis and how disrupted adhesion can contribute to developmental abnormalities and disease.

Integrins bind ligands and transmit information from outside the cell to inside or vice-versa through an "outside-in signaling" or "inside-out signaling." In "outside-in signaling," external factors in the extracellular space bind to exposed ligand binding sites on integrins. This causes the inactive protein to undergo a conformational change to become active. Integrins are often clustered on the cell membrane. Repetitive and regularly spaced ligand binding events provide an effective stimulus.

Here, we present a protocol for the isolation of increasingly insoluble/aggregated alpha-synuclein (α-syn) from post-mortem human brain tissue. Through the utilization of buffers with increasing detergent strength and high-speed ultracentrifugation techniques, the variable properties of α-syn aggregation in diseased and non-diseased tissue can be examined.
