Msc Characterization Protocol

An MSC characterization protocol is a standardized workflow for identifying and validating mesenchymal stem cells, which are multipotent stromal cells relevant to tissue development and repair. The process typically combines assessment of cell morphology and growth with surface-marker analysis, often by flow cytometry, and functional tests that evaluate differentiation toward osteogenic, adipogenic, or chondrogenic lineages under defined culture conditions. In developmental biology, these measurements help distinguish MSC populations from other progenitor cells, assess developmental potential, and compare cells isolated from different tissues or experimental treatments. Consistent characterization also improves reproducibility in studies of lineage commitment, tissue formation, and regenerative biology.

MitoCeption: Transferring Isolated Human MSC Mitochondria to Glioblastoma Stem Cells

Here, a protocol (MitoCeption) is presented to transfer mitochondria, isolated from human mesenchymal stem cells (MSC), to glioblastoma stem cells (GSC), with the goal of studying their biological effects on GSC metabolism and functions. A similar protocol can be adapted to transfer mitochondria between other cell types.

Herein, we communicate protocols to efficiently generate and administer therapeutic spherical aggregates or 'spheroids' of MSCs primed under xeno-free conditions for experimental and clinical applications.

This video describes the technique of seeding mesenchymal stem cells (MSCs) onto a biodegradable poly(lactic acid) (PLA) scaffold. The prepared scaffolds can be implanted into the surgical resection cavity of the brain from where they may migrate towards tumor foci. Further, these cells can be engineered to express therapeutic proteins to stimulate apoptosis in cancer cells.
