Water Volume Method

The Water Volume Method is a quantitative approach for determining how much water is present, collected, stored, or transferred in an environmental system. It works by measuring water with calibrated containers or instruments, or by calculating volume from dimensions, depth, flow rate, and collection time under defined conditions. Researchers and environmental professionals use these measurements to assess stream discharge, runoff, storage capacity, water use, and changes in aquatic or terrestrial systems. Reliable volume estimates support watershed monitoring, resource management, pollution studies, and comparisons of environmental conditions across sites and sampling periods.

A Small Volume Procedure for Viral Concentration from Water

An approach was developed for identifying optimal viral concentration conditions for small volume water samples using spikes of human adenovirus. The techniques described here are used to identify concentration parameters for other viral targets, and applied to large-scale viral concentration experimentation.

The goal of this procedure is to demonstrate the reproducibility and adaptability of using a microtiter plate format for microalgal screening. This rapid screen combines WATER-Pulse-Amplitude-Modulated (WATER-PAM) fluorometry to measure photosynthetic yield as an indicator of Photosystem II (PSII) health with small volume bacterial-algal co-cultures.

A protocol involving integrated concentration, enrichment, and end-point colorimetric detection of foodborne pathogens in large volumes of agricultural water is presented here. Water is filtered through Modified Moore Swabs (MMS), enriched with selective or non-selective media, and detection is performed using paper-based analytical devices (µPAD) imbedded with bacterial-indicative colorimetric substrates.
