Oral Raav Delivery

Oral rAAV delivery is a noninvasive gene-transfer approach that administers recombinant adeno-associated virus vectors through the gastrointestinal tract, offering a potential route for genetic intervention without injection. After oral administration, the vector must survive the digestive environment, reach intestinal tissues, and enter target cells, where its capsid releases a therapeutic gene that can support episomal expression. Research focuses on capsid selection, formulation, and protection from degradation to improve uptake and tissue specificity. In genetics, oral rAAV delivery may enable treatment of intestinal disorders, localized protein production, and broader systemic gene delivery, while addressing key barriers in vector stability and transduction efficiency.

The following protocol describes a novel method for chronic oral drug administration using an orally dissolving strip (ODS) in lieu of the more commonly used oral gavage method. We demonstrate that preclinical, oral drug delivery using the ODS method represents a safe, convenient, and humane alternative to oral gavage.
