Uv-crosslinking Independent

UV-crosslinking-independent methods are experimental approaches for studying biomolecular interactions without using ultraviolet light to create covalent links, an important option in genetic and molecular biology research. These methods typically preserve RNA-protein or DNA-protein complexes through native immunoprecipitation or use chemical crosslinkers, such as formaldehyde, to stabilize molecules that are physically close before purification and sequencing. By avoiding UV exposure, they can reduce dependence on ultraviolet-sensitive interactions and specialized irradiation conditions. Applications include mapping regulatory protein binding, characterizing ribonucleoprotein complexes, and identifying interactions that influence transcription, RNA processing, translation, and genome regulation.

Crosslinking Neuronal Surface Proteins in the Mouse Brain using a Chemical Crosslinking Assay

This video demonstrates the crosslinking of neuronal surface proteins using Bis-sulfosuccinimidyl suberate or BS3. Isolated and chilled mouse brain is sectioned coronally. The desired region is extracted from the sections, minced, and treated with BS3. BS3 crosslinks surface proteins like GABA receptors, leaving internal proteins unaltered. The reaction is quenched with glycine before processing the samples for further analysis.

Nanosponge Tunability in Size and Crosslinking Density

This article describes a process for tuning the size and crosslinking density of covalently crosslinked nanoparticles from linear polyesters containing pendant functionality. By tailoring synthesis parameters (polymer molecular weight, pendant functionality incorporation, and crosslinker equivalents), a desired nanoparticle size and crosslinking density can be achieved for drug delivery applications.
