Endogenous Locus Expression

Endogenous locus expression is the production of a gene or engineered gene product from its native position within the genome, preserving the surrounding regulatory context. Because transcription remains governed by the locus’s promoters, enhancers, chromatin state, and cell-specific signals, expression levels and timing more closely reflect physiological regulation than many ectopic systems. In immunology and infection research, this approach can introduce fluorescent tags, reporters, or defined mutations into immune or pathogen-response genes while retaining endogenous control. It supports accurate analysis of gene activation, protein localization, host-pathogen interactions, and cellular responses, helping researchers distinguish effects caused by normal regulation from artifacts of overexpression.

We provide a method for testing BRCA1 variants in a tissue culture based assay for homologous recombination repair of DNA damage by depleting endogenous BRCA1 protein from a cell using RNAi and replacing it with a BRCA1 point mutant that contains a coding change.
