Pathogen Entry Assessment

Pathogen entry assessment is the systematic evaluation of how an infectious agent attaches to, penetrates, or is internalized by host cells, a critical step in establishing infection. In immunology and infection research, investigators compare entry under defined conditions using cell-based assays, microscopy, or reporter readouts, often with receptor-blocking treatments and temperature or time controls to distinguish binding from internalization. These measurements can identify host receptors, uptake routes, cellular tropism, and barriers to infection. The resulting evidence supports studies of pathogenesis, host defense, antimicrobial or antiviral development, and strategies that prevent pathogens from accessing susceptible cells.

Here, we describe a methodology to obtain a pure population of human myoblasts from adult muscle tissue. These cells are used to study in vitro skeletal muscle differentiation and, in particular, to study proteins involved in Ca2+ signaling.

Assessing Pathogenic Bacteria–Induced Mortality in C. elegans Larvae

Source: Anderson, Q. L., et al. A High-throughput, High-content, Liquid-based C. elegans Pathosystem. J. Vis. Exp. (2018)The video demonstrates a liquid-based assay where genetically modified C. elegans larvae are exposed to pathogenic bacteria in a microplate. Bacterial toxins secreted into the medium induce host stress and death. A fluorescent dye stains dead worms, allowing imaging-based quantification of mortality to assess genetic susceptibility or resistance.

(2025)This video demonstrates the quantification of intracellular survival of a methicillin-resistant bacterial pathogen in mouse peritoneal macrophages. It outlines the steps involved in infection, extracellular bacterial clearance, host cell lysis, and colony enumeration to assess bacterial survival within host macrophages.

Adjusting Entries

2025

In accounting, a business's economic activities are segmented into designated time intervals, typically monthly, quarterly, or annually, known as accounting periods. This segmentation facilitates consistent tracking, summarization, and reporting of financial data, enabling stakeholders to accurately evaluate a company's performance and financial position. Companies must incorporate adjusting entries at the close of each period to ensure that financial reports conform to the accrual basis of...
