Bone Marrow Progenitor Cells

Bone marrow progenitor cells are immature blood-forming cells that can develop into specific lineages of mature blood cells, making them important for maintaining blood production and studying disease. They arise from hematopoietic stem cells and respond to signaling cues within the bone marrow microenvironment, which regulate their proliferation, survival, and differentiation into myeloid or lymphoid cells. In medicine, these cells help researchers understand hematopoiesis, investigate blood disorders, and evaluate treatments that affect bone marrow function. Their biology also supports the clinical use of bone marrow transplantation and the development of regenerative strategies for restoring blood-cell production.

Here we describe an optimized, highly reproducible protocol to isolate Mesodermal Progenitor Cells (MPCs) from human bone marrow (hBM). MPCs were characterized by flow cytometry and nestin expression. They showed the ability to give rise to exponentially growing MSC-like cell cultures while retaining their angiogenic potential.

The nature of the interactions between hematopoietic stem and progenitor cells (HSPCs) and bone marrow niches is poorly understood. Custom hardware modifications and a multi-step acquisition protocol allow the use of two-photon and confocal microscopy to image ex vivo labeled HSPCs homed within bone marrow areas, tracking interactions and movement.
