Nitrite Nitrate Analysis

Nitrite and nitrate analysis is the quantitative measurement of these nitrogen-containing anions in biological samples, helping assess nitric oxide metabolism and related physiological or pathological changes. In a common colorimetric workflow, nitrate is first reduced to nitrite, which reacts with sulfanilamide and N-(1-naphthyl)ethylenediamine to form a colored azo compound measured by spectrophotometry; chromatography and electrochemical detection provide complementary approaches. In medicine, these measurements support studies of endothelial function, cardiovascular biology, inflammation, and dietary nitrate exposure, while careful sample handling improves analytical reliability and interpretation.

Preparation of Rat Skeletal Muscle Homogenates for Nitrate and Nitrite Measurements

We present protocols for three different methods for the homogenization of four different muscle groups of rat skeletal muscle tissue to measure and compare the levels of nitrate and nitrite. Furthermore, we compare different sample weights to investigate whether tissue sample size affects the results of homogenization.

Zebrafish as a Model to Assess the Teratogenic Potential of Nitrite

Exposure to teratogens may cause birth defects. Zebrafish are useful for determining the teratogenic potential of chemicals. We demonstrate the utility of zebrafish by exposing embryos to various levels of nitrite and also at different times of exposure. We show that nitrite can be toxic and cause severe developmental defects.

Nitric oxide (NO) is an important signaling molecule in vascular homeostasis. NO production in vivo is too low for direct measurement. Chemiluminescence provides useful insight into NO cycle via measuring its precursors and oxidation products, nitrite and nitrate. Nitrite / nitrate determination in body tissues and fluids is explained.
