Fluorogenic Reporter

A fluorogenic reporter is a molecular probe that produces a measurable fluorescent signal only after a defined biochemical or chemical event, making it useful for detecting otherwise difficult-to-visualize processes. In many designs, a dark or quenched fluorophore becomes fluorescent when an enzyme cleaves a masking group or when a target-induced reaction changes its structure, so signal intensity reports activity or localization. In neuroscience, fluorogenic reporters can help monitor neuronal signaling, enzyme function, and dynamic molecular events in living cells and tissues, while low background improves spatial contrast and quantitative analysis.

This video demonstrates an assay to screen for the proteolytic activity of proteases using fluorogenic peptides. The protease recognizes its cleavage site on the peptide, cleaving it and separating the quencher from the fluorophore, enabling its fluorescence emission. The fluorescence signal is detected and analyzed to check for the cleavage efficiency of different peptide variants.
