Ischemic Injury

Ischemic injury is tissue damage caused by inadequate blood flow, which deprives cells of oxygen and glucose and is a major mechanism of brain damage after stroke, cardiac arrest, or vascular occlusion. In nervous tissue, energy failure reduces ATP production, disrupts ion gradients, and impairs membrane transport, leading to glutamate excitotoxicity, calcium overload, oxidative stress, and neuronal cell death. Experimental ischemic injury models help researchers examine the progression from reversible dysfunction to irreversible damage, evaluate neuroprotective strategies, and identify therapeutic targets. Understanding these mechanisms supports improved approaches to stroke treatment, recovery, and prevention of long-term neurological impairment.

2025

The video demonstrates the procedure for creating a mouse model to study neonatal hypoxic-ischemic brain injury. It outlines the steps for surgically inducing ischemia and subjecting the animal to hypoxia to exacerbate brain damage. Finally, it shows how to assess the resulting brain lesions, ensuring the mouse model is prepared for further scientific investigation.

We performed unilateral carotid artery occlusion on postnatal day 7-10 CD-1 mouse pups to create a neonatal hypoxic-ischemic (HI) model and investigated the effects of HI brain injury. We studied neurobehavioral functions in these mice compared to non-operated normal mice.

Compensatory Limb Use and Behavioral Assessment of Motor Skill Learning Following Sensorimotor Cortex Injury in a Mouse Model of Ischemic Stroke

Mouse models have become increasingly popular in studies of behavioral neuroscience. As models advance, it is important to develop sensitive behavioral measures specific to the mouse. This protocol describes the Pasta Matrix Reaching Task, which is a skilled motor task for use in mouse models of stroke.

The postnatal rat model for hypoxic-ischemic brain injury is a well-established model of human neonatal hypoxic ischemic encephalopathy (HIE). In this article, we describe the model of HIE in post-natal rat pups.

The goal of this protocol is to photochemically induce ischemic injury to the posterior optic nerve in rat. This model is critical to studies of the pathophysiology of posterior ischemic optic neuropathy, and therapeutic approaches for this and other optic neuropathies, as well as of other CNS ischemic diseases.
