Light Microscopy Visualization

Light microscopy visualization is the use of visible light, optical lenses, and image capture to magnify and resolve structures that cannot be seen with the unaided eye. In neuroscience, transmitted or reflected light creates contrast through differences in absorption and scattering, while fluorescence microscopy uses labeled molecules that emit light after excitation, allowing selected cells or proteins to be distinguished. These approaches reveal neuronal morphology, synaptic organization, and cellular interactions in cultured samples, tissue sections, and living preparations. By linking structure with location and activity, light microscopy supports studies of brain development, neural circuits, disease mechanisms, and responses to experimental treatments.

Here, we describe a light-sheet microscopy approach to visualize the cardiac CD45+ leukocyte infiltrate in a murine model of aseptic fulminant myocarditis, which is induced by the intratracheal diphtheria toxin treatment of CD11c.DTR mice.

The light microscope is an instrument used by researchers in many different fields to magnify specimens to as much as a thousand times their original size. In its simplest form, it is composed of a clear lens that magnifies the sample and a light source to illuminate it. However, most light microscopes are much more complex and house numerous fine-tuned lenses with tightly controlled dimensions all within the body of the microscope itself and in components such as the objectives and eyepieces.

We demonstrate the fabrication of a low-cost cryogenic stage designed to fit most reflected light microscopes. This lab-built cryogenic stage enables efficient and reliable correlative imaging between cryo-light and cryo-electron microscopy.

Lattice Light Sheet Microscopy to Visualize Receptor-Ligand Interactions in Live Cells

In this video, we describe the lattice light-sheet microscopy technique to visualize interactions between live T-cells and antigen-presenting cells. The method involves focusing an ultrathin lattice light sheet on an interacting cell pair to obtain a series of two-dimensional images, which are combined to obtain high-resolution four-dimensional images that reveal the spatiotemporal dynamics of the cell-cell interaction.

Correlative Light- and Electron Microscopy Using Quantum Dot Nanoparticles

A method is described whereby quantum dot (QD) nanoparticles can be used for correlative immunocytochemical studies of epoxy embedded human pathology tissue. We employ commercial antibody fragment conjugated QDs that are visualized by widefield fluorescence light microscopy and transmission electron microscopy.
