Retinal Thickness Measurement

Retinal thickness measurement is a quantitative assessment of the thickness of retinal tissue, providing a noninvasive indicator of structural changes in the neural retina. In neuroscience, it is commonly performed with optical coherence tomography, which uses reflected light and interferometric imaging to generate cross-sectional retinal scans; software then identifies anatomical layer boundaries and calculates thickness. Measurements can be compared across retinal regions, individuals, or time points to evaluate tissue loss, swelling, or treatment-related change. This approach supports research on retinal and optic nerve disorders, neurodegenerative disease, and brain-eye relationships by linking microscopic structural alterations with neural function and disease progression.

Here we describe a cost-effective technique for organotypic culture of adult porcine retina for seven days. Briefly, a sterile filter paper was used to lift the neural retina off from the RPE and place photoreceptor side up on an insert raised by a custom-made stand.
