Volume Fraction

Volume fraction is the proportion of a material or structural component’s volume relative to the total volume of a defined sample, providing a standardized measure of composition. In neuroscience, it is calculated by identifying and segmenting tissue compartments, such as gray matter, white matter, blood vessels, cells, or extracellular space, and dividing each component’s volume by the reference tissue volume. Imaging, histology, and stereological methods can estimate these proportions across brain regions or experimental conditions. Comparing volume fractions helps researchers characterize tissue organization, detect structural changes, and relate microscopic composition to brain function, development, disease, or treatment response.

This protocol describes real-time iontophoresis, a method that measures physical parameters of the extracellular space (ECS) of living brains. The diffusion of an inert molecule released into the ECS is used to calculate the ECS volume fraction and tortuosity. It is ideal for studying acute reversible changes to brain ECS.

Fractional Distillation

Source: Laboratory of Dr. Nicholas Leadbeater — University of Connecticut Distillation is perhaps the most common laboratory technique employed by chemists for the purification of organic liquids. Compounds in a mixture with different boiling points separate into individual components when the mixture is carefully distilled. The two main types of distillation are "simple distillation" and "fractional distillation", and both are widely used in organic chemistry laboratories. Simple distillation...

The accompanying video describes the use of the GELFREE 8100 Fractionation System, which partitions complex protein samples on the basis of molecular weight and recovers the fractions in liquid phase. The video describes how the technology works, how it is used, and provides resultant data, with polyacrylamide gel electrophoresis analysis of fractionated bovine liver homogenate.

Constant Volume Calorimetry

2020

Calorimeters are useful to determine the heat released or absorbed by a chemical reaction. Coffee cup calorimeters are designed to operate at constant (atmospheric) pressure and are convenient to measure heat flow (or enthalpy change) accompanying processes that occur in solution at constant pressure. A different type of calorimeter that operates at constant volume, colloquially known as a bomb calorimeter, is used to measure the energy produced by reactions that yield large amounts of heat and...
