X線品質の結晶は、一般に対称的な形状を形成し、滑らかで光を反射する面を持っています。偏光顕微鏡で見ると透明になりますが、90度回転させるとほとんどは暗くなるはずです。これは、高度に順序付けられた構造を示します。これらの結晶を成長させるために、液液拡散がよく使用されます。これは、2つの混和性溶媒を使用します:低密度溶媒、または沈殿剤で、再結晶する化合物が不溶性です。化合物が可溶性である高密度溶媒。通常、沈殿物と溶媒の体積比は2:1です。

まず、テキストプロトコルにある必要な機器を入手します。化合物用の溶媒と密度の低い沈殿物を入手します。

次に、使用済みのピペットフィルターを取り外し、2つ目のフィルターをNMRチューブに入れます。約1.5mLの沈殿物をチューブにピペットで入れます。溶媒が重力によってフィルターを通過するのを待ちます。これからは、操作中にフィルターを乱さないように注意してください。すべての沈殿物がチューブにろ過されたら、フィルターを取り外し、チューブにキャップをします。キャビネットなど、攪拌されない場所に置いてください。
少なくとも1日後、チューブの結晶成長を検査します。結晶が存在しない場合、または結晶が非常に小さい場合は、サンプルチューブを邪魔せずに放置してください。結晶が見える場合は、溶媒層を乱さずに結晶の大きさや形状を確認してください。
結晶が大きく、明確に定義され、クラスター化されていない場合は、顕微鏡で結晶を検査して、X線品質になる可能性を確認します。回折計がスキャンを開始する準備ができるまで、チューブから結晶を取り出さないでください。結晶構造に溶媒分子が組み込まれている場合、結晶を乾燥させると結晶が劣化します。X線結晶構造解析を用いて、この暗赤色紫色の結晶の分子構造がテトラフェニルポルフィリンであることが確認されました。
X線結晶構造解析は、化学および生化学において不可欠な分析ツールです。
再結晶方法には、加熱と冷却、液液拡散、蒸気拡散、およびゆっくりとした蒸発が含まれます。単一溶媒系のゆっくりとした蒸発では、化合物を少量の溶媒に溶解し、キャップに小さな穴が開いた容器に入れます。溶媒が蒸発すると、化合物が結晶化し始めるまで濃度が増加します。
タンパク質の機能性は、多くの場合、その構造に関連しています。しかし、タンパク質は結晶化が非常に難しい場合があります。X線品質のタンパク質結晶を成長させるには、専門的な技術を開発する必要があります。ここでは、一滴のタンパク質溶液を一滴の沈殿物と混合し、この混合物を純粋な沈殿物の入ったチャンバーに密封します。溶媒蒸気が液滴から拡散すると、液滴中のタンパク質の溶解度が低下し、タンパク質はゆっくりと結晶化します。別の技術は、タンパク質溶液と沈殿物を鉱油の下に混合します。これらの技術を用いて、様々なタンパク質を結晶化して分析することができます。

This ordered structure is essential because crystal quality directly influences the accuracy of structural determination in X-ray crystallography.

Rapid crystal formation causes molecular disorder, incorporates impurities, or creates fused crystals instead of single crystals. The pipette filter removes solid impurities from the compound solution before crystallization begins. A small piece of Kimwipe is placed in the glass pipette and pressed to the bottom. The sample solution passes through the filter by gentle bulb pressure, ensuring only pure dissolved compound enters the NMR tube for clean crystal formation.

When X-rays hit a crystal with ordered atoms, electrons scatter the rays, producing constructive interference and a characteristic diffraction pattern. Protein crystallization often requires specialized techniques like purifying compounds by recrystallization under mineral oil.

Single-crystal X-ray diffraction preserves three-dimensional structural information because each crystal orientation is distinct.
