次に、剣状突起から恥骨への正中線切開を行い、マイクロリトラクターを使用して腹腔を開きます。蚊の鉗子を使用して剣状突起をマウスの頭に向かって引っ込め、柔らかい粘土を使用して鉗子と剣状突起をこの位置に保持します。1〜2ミリリットルの滅菌済み37°CPBSで腹腔を水分補給し、濡れた綿棒を使用して小腸をマウスの頭に向かって穏やかに動かします。
組織を傷つけないように小腸と後腹膜の間の靭帯を切断し、濡れた不織布ガーゼスポンジで小腸と大腸を包みます。腸組織を腹腔の尾側に置き、小さな濡れた不織布ガーゼスポンジを使用して、肝臓全体を剣状突起に向かって穏やかに引っ込め、肝門と肝十二指腸靭帯を露出させます。肝ハイラム、門脈の腹側にある総胆管、および肝ハイラムで主胆道合流点に排出される左右の肝胆管を見つけます。
10-0ナイロン縫合糸と縫合針の鈍い面を使用して、左肝胆管葉を結紮します。縫合糸が埋入されたら、小腸と大腸を解剖学的位置に戻し、腸のねじれや腹腔内出血がないことを確認します。次に、蚊の鉗子を取り外し、連続した4-0 Vicryl縫合糸で筋肉と皮膚の層を別々に閉じ、マウスを完全に回復するまで監視しながらマウスをヒートランプの下に置きます。
不溶性肝タンパク質凝集のための部分胆管結紮マウスと完全胆管結紮マウスとの間の血清肝生化学の比較は、部分胆管結紮マウスにおける最小限の肝障害または胆汁うっ滞、および完全胆管結紮トランスジェニック動物における重篤な損傷および酵素蓄積を明らかにする。部分胆管結紮術を受けたマウスは、時間の経過とともに葉特異的閉塞性胆汁うっ滞を発症したため、手術後14日で結紮された葉は、正常な暗赤褐色を維持していた結紮されていない葉と比較してわずかに色が薄く見えます。結紮された葉はまた、結紮によって誘発される限局性胆汁うっ血に応答して、結紮されていない葉と比較して胆管の増殖と線維化の増加を示します。
