現在、桿体細胞下区画を単離することを目的としたいくつかの方法がある。しかしながら、これらの方法は、長くて再現が困難であり得るか、またはかなりの量の網膜分離物を必要とする。ロッドアウターセグメント(ROS)調製物は、密度勾配遠心分離19を介して、例えば、網膜ホモジネートからROSを分離するために一般的に使用される。この方法はウェスタンブロットに広く使用されているが、この手順は非常に時間がかかり、最低8〜12匹のマウス網膜20を必要とする。一方、凍結マウスおよびラット網膜の直列接線切片化は、OS、IS、CB、およびST9、11、13を単離する際に首尾よく実施されている。しかし、この方法は、接線切除の前に網膜層を整列させるために、小さくて高度に湾曲したマウス網膜を完全に平坦化する必要があるため、技術的に困難である。視覚系の疾患を再現するマウスモデルやトランスジェニックマウスは数多く存在するため、個々の桿体コンパートメントを確実に、迅速に、そして容易に分離する技術の創出は、各特殊なコンパートメントで起こる生理学的プロセスと、健康および疾患における視覚プロセスの根底にあるメカニズムを明らかにする上で有望である。
これらの調査を容易にするために、我々は、現在のプロトコルよりも容易に桿体細胞下区画を分離する2つの剥離方法を説明する。第1の剥離方法は、パッチクランプ記録21のために蛍光標識されたバイポーラ細胞を露出させる技術から適合し、セルロース濾紙を使用して、生きた単離されたマウス網膜からROSを順次除去する(図1)。第2の方法は、chick22およびfrog23網膜から3つの一次網膜細胞層を単離する手順から適合し、粘着テープを使用して凍結乾燥網膜からROSおよび桿体内部セグメント(RIS)を除去する(図2)。どちらの手順も1時間で完了でき、かなりユーザーフレンドリーです。我々は、C57BL/6Jマウスからの暗適応および光曝露網膜を利用して、ロッドトランスデューシン(GNAT1)およびアレスチン(ARR1)の光誘発転座を実証することによって、ウェスタンブロットに対するこれら2つの分離プロトコルの有効性の検証を提供する。さらに、テープ剥離法を用いて、我々の技術が免疫細胞化学(ICC)によって取得されたタンパク質局在データとウェスタンブロットとの間の不整合を調べ、対処するために使用することができるという追加の証拠を提供する。具体的には、我々の技術は、1)プロテインキナーゼC-α(PKCα)アイソフォームが双極性細胞だけでなく、マウスROSおよびRISにも低濃度ではあるが24,25存在し、2)ロドプシンキナーゼ(GRK1)が単離されたOSサンプル中に主に存在することを示した。これらのデータは、特定のロッドタンパク質とレチナールタンパク質を分離および定量するための2つの剥離技術の有効性を示しています。

Two Peeling Methods for the Isolation of Photoreceptor Cell Compartments in the Mouse Retina for Protein Analysis
