Sampling Cerebrospinal Fluid and Blood from Lateral Tail Vein in Rats During EEG Recordings

体液の組成は多くの生理学的および病理学的動態を反映しているため、ホルモン、成長因子、タンパク質、低分子ノンコーディングRNAなどの目的の分子を測定するために、多くの実験状況で生体液体サンプルが一般的に採取されます。具体例としては、てんかんのバイオマーカー研究における生体液のサンプリングがあります。これらの研究では、脳脊髄液(CSF)と血漿中の分子のレベルを比較することが望ましいです。てんかん動物におけるビデオ脳波モニタリングと組み合わせたCSFと血漿サンプリングの組み合わせは、推定診断および予後バイオマーカーの検証のための有望なアプローチです。ここでは、てんかんラットの大槽からのCSF離脱と側尾静脈からの採血を組み合わせた手順を、ビデオ脳波を継続的にモニターする手順について説明します。この手順には、一般的に使用される他の手法に比べて大きな利点があります。痛みや侵襲性を最小限に抑え、麻酔時間を短縮した迅速なサンプリングを可能にします。さらに、テザーおよびテレメトリーEEG記録ラットの両方でCSFおよび血漿サンプルを取得するために使用でき、数日間の実験で繰り返し使用できます。イソフルラン麻酔を短くすることでサンプリングによるストレスを最小限に抑えることで、生体液中の調査分子の真のレベルをより正確に反映する測定が期待されます。適切な分析アッセイの利用可能性に応じて、この技術を使用して、同時に脳波記録を実行しながら、複数の異なる分子のレベルを測定することができます。

SE後1か月で電極を埋め込んだ9匹の対照ラットと18匹の慢性てんかんラットで実施されたさまざまなCSFおよび採血手順の結果が、成功率の観点から報告されています。移植後、すべてのラットを1か月間ビデオ脳波でモニターし、その間、実験の最後の2週間(すなわち、SE後52、55、58、61、および64日目)に3日ごとにCSFと血液を5回回収しました。異なる動物における複数の離脱からのデータを使用して、双頭インプラントを授造したラット(CSF離脱のためにカニューレ挿入)におけるCSF採取の成功率と、係留またはテレメトリー電極のみでCSF採取の成功率(システルナマグナ穿刺によって実施)を比較しました(表1)。異なる動物において、真空採血または尾搾乳が血漿サンプルの品質に与える影響を評価しました(表2)。この目的のために、遊離ヘモグロビンの検出に414nmの紫外分光光度法分析が使用されました。統計解析には、市販のソフトウェアを使用し、Kruskal-Wallisまたは事後テューキーの多重比較検定による一元配置分散分析を使用しました(p&lt;0.05は統計的に有意であると見なされます...
