摂氏4度で2時間インキュベートします。次に、抗原をロードしたビーズを含む細胞を、蓋を開けた状態で磁気ラックに置きます。蓋の中にある液体をチューブにピペットで入れます。
2分間待ってから、バインドされていないセルを破棄します。最初の抗原ビーズの選択について説明したように、残りのすべてのステップを実行します。3回のネガティブ選択と1回のポジティブ選択の後、細胞を1ミリリットルのSDCAAに急速に再懸濁し、大量のSDCAAを含むシェーカーフラスコに移します。
酵母ライブラリーのSGCAAでタンパク質の表面発現を一晩誘導した後、多様性の10倍をカバーするのに十分な細胞をペレット化し、上清を廃棄します。ペレットをPBSAに再懸濁し、マイクロ遠心チューブに移します。各チューブで3,000万個の細胞を染色します。
抗原を含まないコントロールチューブ1本を含め、多様性に基づいて必要な数のチューブを準備します。次に、チューブを2000 x gで室温で5分間遠心分離します。ペレットを抗原を含む200マイクロリットルのPBSAに再懸濁し、摂氏4度で1時間インキュベートします。
再度遠心分離を行った後、ペレットからPBSAを除去します。次に、ディスプレイ染色および抗原検出のために、100 μリットルの低温PBSA含有抗体に細胞を再懸濁します。摂氏4度で30分間インキュベートします。
インキュベーション後、細胞を2000 x gで4°Cで5分間遠心分離します。ペレットに1ミリリットルのPBSAを加え、遠心分離を繰り返します。次に、ペレットが乾燥するのを防ぐために、上澄みの大部分を取り除き、20〜30マイクロリットルだけを残します。
選別する直前に、ペレットを冷たいPBSAに再懸濁します。次に、細胞を直接SDCAA培地に選別します。選別後、細胞を大量のSDCAA培地を含むシェーカーフラスコに移し、摂氏30度でインキュベートし、180RPMで振とうします。
