1. サンプルと材料
<ol>
	<li>青海西藏高原からさまざまな カバ のシーバックソーンサンプルを収集します。 注:サンプルは、成都中医薬大学のYue Liu准教授によって同定されました。バウチャー標本は成都中医薬大学民族医学部に寄託されました。サンプリング情報を 表1に示します。この研究で使用した試薬と機器の詳細は 、材料表に記載されています。</li>
	<li>サンプルを40°Cのオーブンで72時間乾燥させます。</li>
	<li>各シーバックソーンサンプル20gを粉末に粉砕し、3番ふるいに通します。各サンプルを3回並列反復で処理します。ふるいにかけられなかったサンプルを粉砕し続け、すべてふるいにかけられるまでふるいにかけます。</li>
	<li>フォローアップ研究のためにサンプルを-20°Cで保存します。</li>
</ol>
2. シーバックソーン多糖類の抽出
注:この研究では、多糖類30を得るためにいくつかの変更を加えたシーバックソーンの以前に確立された抽出方法を採用しました。
<ol>
	<li>5.0 gのシーバックソーンサンプル粉末を秤量し、250 mLの丸底フラスコに入れます。粉末に50 mLの無水エタノールを加え、室温(RT;25°C)で24時間浸して親油性成分を除去します。各サンプルを3回並列反復で処理します。</li>
	<li>一晩浸したサンプル粉末をブフナー漏斗を使用してろ過し、濾液を廃棄してフィルター残留物を取得します。フィルター残渣を50mLの脱イオン水で3回洗浄し、40°Cで一定の重量に乾燥させます。</li>
	<li>脱脂フィルター残渣を乾燥した250 mLの丸底フラスコに移し、150 mLの脱イオン水を固液比1:30(g/mL)で加え、80°Cのウォーターバスで2回、毎回2時間還流抽出します。濾液を分離して、粗海クロウメモドキ多糖類溶液を得る。</li>
	<li>ロータリーエバポレーターを使用して50°Cで1.5時間抽出物を濃縮します。濃縮溶液は、後で使用するために4°Cの冷蔵庫に保管してください。</li>
	<li>Sevage試薬(CHCl 3:C4H9OH = 4:1、v:v)を粗多糖類溶液に、Sevage試薬:粗多糖類溶液の体積比1:4で添加します。マグネチックスターラーで混合物を1050rpmの速度で20分間攪拌します。</li>
	<li>液体を50mL遠心分離管に移し、4000 × g で15分間遠心分離し、上清を回収します。2つの相の間の界面に白い凝集沈殿物が現れないまで、上記の操作を3回繰り返します。</li>
	<li>タンパク質除去後のシーバックソーン多糖類抽出物を濃縮します。無水エタノール沈殿のために無水エタノールを体積比1:3で加え、混合物を4°Cで一晩置きます。</li>
	<li>無水エタノールの上層を取り除き、混合物を50mL遠心分離管に移し、4000 × g で15分間遠心分離して沈殿物を得る。沈殿物を凍結乾燥して、総多糖類を得る。</li>
	<li>以下の式(1)を使用して、総多糖類収量を計算します。

3. 多糖類含有量の測定
<ol>
	<li>溶液の調製<ol>
		<li>フェノール試薬溶液の調製<ol>
			<li>フェノール3gを正確に計量し、茶色のメスフラスコに移します。50 mLの脱イオン水を加えてフェノールを溶解します。</li>
			<li>均一になるまで混合物を完全に振ってください。フラスコにラベルを付けて、後で使用するために保管します。</li>
		</ol>
		</li>
		<li>グルコース標準溶液の調製<ol>
			<li>10 mgの無水ブドウ糖を正確に計量し、10 mLのメスフラスコに移します。脱イオン水を加えてブドウ糖を溶解し、混合物をよく振ってください。</li>
			<li>混合物を脱イオン水で所望の体積に希釈して、グルコース標準溶液を得る。</li>
		</ol>
		</li>
		<li>試験サンプル溶液の調製。<ol>
			<li>凍結乾燥シーバックソーン多糖類(ZG1-ZG5およびJZ1-JZ5)5 mgをメスフラスコ(n = 10)に正確に計量します。脱イオン水を各フラスコに加え、最終容量は50 mLになります。</li>

	</li>
	<li>標準曲線の構築 注:フェノール-硫酸法では、多糖類が濃硫酸と反応してフルフラール誘導体を生成し、フェノールと縮合してオレンジがかった黄色の化合物を形成します。この化合物の最大吸収波長は約490nmです。したがって、この実験の検出波長は490nm22に設定されました。<ol>
		<li>標準グルコース溶液を脱イオン水で0.1 mg / mLの濃度に希釈します。マイクロピペットを使用して、200、300、400、500、600、および700 μLのグルコース標準溶液を6本の別々の清潔な試験管に慎重に移します。総量1.0mLまで純水を加え、各溶液を穏やかに混合して均一に希釈します。</li>
		<li>上記の試験管に1.0mLの6%フェノール溶液と5.0mLの硫酸を加えます。よく混ぜて冷ましてから、RTで30分間放置します。 注:フェノールは有毒な化合物であり、濃硫酸は腐食性があります。フェノールおよび硫酸溶液を調製するときは手袋を着用し、手順はヒュームフード内で行う必要があります。</li>
		<li>前述の試料をキュベットに移し、試料の代わりに純水をブランクとして使用する。490 nmでの吸光度をUV-Vis分光光度計と3回の反復で測定します。</li>
		<li>グルコース濃度をx軸、吸光度値をy軸とする標準曲線をプロットします。回帰式を取得し、相関係数(R²)を計算します。 注:UV-Vis分光光度計は、30分前に予熱する必要があります。このステップで吸光度を測定する場合、吸光度は0.2〜0.8でなければなりません。したがって、サンプルの濃度を定量化する必要があります。</li>
	</ol>
	</li>
	<li>方法論研究<ol>

4. DPPHラジカル消去活性31,32
<ol>
	<li>2.7 mgのDPPHを空の50 mLメスフラスコに正確に計量します。それを50 mLの無水エタノールに溶解して、0.054 mg/mLの濃度のDPPHストック溶液を調製し、DPPH作業溶液を得る。 注:DPPH溶液は暗所に保管する必要があります。</li>
	<li>5 mgのビタミンC(Vc)を空の10 mLメスフラスコに正確に計量し、10 mLの脱イオン水に溶解して、0.5 mg/mLの濃度のVc溶液を調製します。連続して希釈して、0.5、0.125、0.03125、0.0078125、および0.001953 mg / mLの濃度のVc試験溶液を調製します。</li>

粗多糖類の収量 多糖類はHWEによって抽出されました。ウォーターバスの還流を補足図1に示します。多糖類抽出物溶液の濃度を補足図2に示します。脱タンパク後の遠心分離成層効果を補足図3に示します。H. rhamnoides subsp. sinensisの5つのバッチの多糖類収量は、0.89%±0.01%、0.78%±0.02%、1.63%±0.03%、1.50%±0.01%、1.12±0.03%で、平均収量は1.18%±0.02%です(表2)。ステップ2.5の5つのバッチの多糖類収量は、0.07%±3.94%±0.06%、3.53%±0.05%、2.63%±0.08%、および2.83%±0.05%であり、平均収量は3.12%±0.06%です(表2)。t検定の結果、2つの種間の多糖類収量に有意差が見られました(P &lt; 0.01)。今回の結果は、H. gyantsens...
