In-vitro Reconstitution of Bacterial Ubiquitination and VCP/p97-mediated Elimination

本プロトコルでは、哺乳類のA549細胞を利用して、ユビキチン化プロセスを開始するために必要な成分を豊富に含む細胞分解液を生成する方法を詳述しています(図1A)。この溶解液は、濃縮状態であれ、UBE1(E1ユビキチン活性化酵素)、UbE2C(E2ユビキチン共役酵素)、Rbx1-Skp1-Cul1-Fbxw7複合体(E3ユビキチンリガーゼ)などの精製成分と置換されていずれも、精製されたユビキチンおよびATPと混合され、細菌の in vitro ユビキチン化を促進します(図1B)).ユビキチン化後のバクテリオリシス応答を評価するために、精製されたVCP/p97およびその補因子UFD1およびNPLOC4を反応に含めます(図1C)。
