Cold Room Electrophoresis

Cold room electrophoresis is an electrophoretic separation technique performed at reduced temperature to help maintain the stability and biological activity of heat-sensitive biomolecules, especially proteins and nucleic acids. An applied electric field drives charged molecules through a gel or other supporting medium, while differences in molecular charge, size, and conformation determine their migration; refrigeration limits heat buildup generated by electrical resistance and reduces sample degradation. In biochemistry, the method supports protein purification, analysis of macromolecular complexes, and separation of labile samples before downstream assays such as activity measurements or immunochemical detection. It is particularly useful when conventional electrophoresis compromises molecular integrity.
