Wheat Grain Analysis

Wheat grain analysis is the systematic examination of a grain’s physical, chemical, and biochemical properties to assess composition, quality, and suitability for food or research applications. In biochemical analysis, researchers homogenize grain samples and measure constituents such as proteins, carbohydrates, lipids, moisture, enzymes, and secondary metabolites using methods including spectroscopy, chromatography, and colorimetric assays. These measurements reveal how genotype, growing conditions, storage, and processing influence grain composition and functional performance. The resulting data support quality control, nutritional evaluation, breeding programs, detection of deterioration or contamination, and the development of wheat products with targeted processing and nutritional characteristics.

The protocol includes an acetonitrile metabolite extraction method and reversed phase liquid chromatography-mass spectrometry methodology, with acquisition in positive and negative electrospray ionization modes.

In this work, we describe the use of the atom-probe tomography technique for studying the grain boundaries of the absorber layer in a CIGS solar cell. A novel approach to prepare the atom probe tips containing the desired grain boundary with a known structure is also presented here.

This paper provides a detailed method to characterize the microstructure of ultra-fine grained and nanocrystalline materials using a scanning electron microscope equipped with a standard electron backscatter diffraction system. Metal alloys and minerals presenting refined microstructures are analyzed using this technique, showing the diversity of its possible applications.

Methods for the manipulation and analysis of NF-κB-dependent adult hippocampal neurogenesis are described. A detailed protocol is presented for a dentate gyrus-dependent behavioral test (termed the spatial pattern separation-Barnes maze) for the investigation of cognitive outcome in mice. This technique should also help enable investigations in other experimental settings.
