Fluorescent Membrane Marker

A fluorescent membrane marker is a dye, fluorescent protein, or other probe that labels cell membranes, enabling researchers to visualize membrane structure, dynamics, and organization. These markers associate with lipid bilayers or membrane proteins and emit light at characteristic wavelengths after excitation, allowing fluorescence microscopy to track membrane boundaries, movement, and remodeling over time. In bioengineering, fluorescent membrane markers support the development and evaluation of engineered cells, lipid-based materials, tissue constructs, and drug-delivery systems. They also help quantify cell morphology, membrane trafficking, fusion, and interactions with biomaterials, providing spatial and temporal information that links membrane behavior to cellular function.

In this video, we demonstrate a fluorescence-detection size-exclusion chromatography technique to screen for green fluorescent protein (GFP)-fused membrane protein stability in Escherichia coli following detergent solubilization. Proteins exhibiting a symmetrical peak in the size-exclusion profile with little or no free GFP indicate purified proteins with minimum degradation and aggregation, which can subsequently be selected for structure-function analysis.
