Resazurin Conversion Assay

The resazurin conversion assay is a rapid method for estimating cellular metabolic activity, providing an indirect measure of cell viability and growth in bioengineering research. Living cells reduce the blue, nonfluorescent dye resazurin to pink, fluorescent resorufin through intracellular redox reactions, so the resulting color or fluorescence signal reflects metabolic activity under defined assay conditions. Researchers use this readout to monitor cell proliferation, evaluate biomaterial and scaffold biocompatibility, assess cytotoxic responses, and compare tissue-engineering strategies. Because the assay is sensitive and compatible with microplate workflows, it supports quantitative analysis while minimizing disruption to cultured cells.

Most bacterial infections produce a biofilm. By virtue of their environment, biofilm associated bacteria are often phenotypically drug resistant. Novel antibacterial molecules that kill bacteria in biofilms are thus a high priority. We establish an assay to quickly screen for antimicrobial compounds that are effective at eradicating biofilms.

The charcoal agar resazurin assay (CARA) is a semi-quantitative, medium-throughput method to assess activity of test agents against mycobacteria that are replicating, non-replicating, or both. The CARA permits rapid evaluation of time- and concentration-dependent activity and identifies parameters to pursue by colony forming unit (CFU) assays.
