Scaffold Recellularization

Scaffold recellularization is a tissue-engineering process that repopulates a biological or synthetic scaffold with living cells to restore structural and functional features of a target tissue. It typically involves preparing a scaffold with suitable architecture and biochemical cues, introducing selected cells under controlled culture conditions, and supporting their adhesion, migration, proliferation, and maturation throughout the construct. In bioengineering, recellularization helps create tissue models and regenerative grafts for studying cell–matrix interactions, evaluating therapies, and repairing damaged tissue. Progress depends on achieving uniform cell distribution, adequate nutrient transport, and integration between newly formed tissue and the scaffold.

Perfusion decellularization is a novel technique to produce whole liver scaffolds that retains the organ's extracellular matrix composition and microarchitecture. Herein, the method of preparing whole organ scaffolds using perfusion decellularization and subsequent repopulation with hepatocytes is described. Functional and transplantable liver grafts can be generated using this technique.
