Vagina Cervix

The vagina and cervix are interconnected components of the female reproductive tract that provide a protective, elastic passage and regulate communication between the uterus and external environment. Their functions depend on specialized epithelial tissues, connective and smooth muscle layers, and cervical mucus, whose composition and viscosity change with hormonal conditions to influence barrier protection, lubrication, and sperm transport. In bioengineering, these structures guide the development of three-dimensional tissue models, biomaterials, and organ-on-chip systems for studying infection, reproductive health, drug delivery, and tissue repair. Such platforms can improve disease modeling and support more precise evaluation of therapies without relying solely on animal studies.

This article describes a protocol for creating a microfluidic vagina-on-a-chip (Vagina Chip) culture device that enables the study of human host interactions with a living vaginal microbiome under microaerophilic conditions. This chip can be used as a tool to investigate vaginal diseases as well as to develop and test potential therapeutic countermeasures.

This protocol demonstrates a reliable and effective technique to isolate primary fibroblasts from either premenopausal or postmenopausal human vaginal tissue. Existing protocols for vaginal fibroblast isolation do not consider the challenges of cell isolation from senescent tissue. Vaginal tissue was obtained from women after pelvic organ prolapse surgery.
