Cell Transplantation Technique

Cell transplantation is a technique in which viable cells are transferred into a recipient to replace damaged cells, restore tissue function, or study cell behavior in a biological system. The procedure typically involves preparing donor or cultured cells, delivering them to a target tissue, and supporting their survival, engraftment, and integration with host cells; immune compatibility can influence outcomes. In biology research, cell transplantation helps investigate tissue repair, regeneration, disease mechanisms, and cell fate. It also supports the development of regenerative therapies and experimental models for evaluating how transplanted cells respond to their surrounding microenvironment.

This work demonstrates a novel approach to assess the proliferation, differentiation, and vessel-forming potential of vascular endothelial stem cells (VESCs) through mammary fat pad transplantation followed by whole-mount tissue preparation for microscopic observation. A lineage tracing strategy to investigate the behavior of VESCs in vivo is also presented.

The murine cervical heart transplantation model is well suited for immunological as well as ischemia reperfusion injury studies. We modified the procedure using a non-suture cuff technique and performed more than 1,000 successful transplants with this approach. Herein, we provide additional details of this technique to supplement the video.
