Endocytic Pathway

The endocytic pathway is a cellular transport system that brings substances from the plasma membrane into the cell, enabling nutrient uptake, membrane maintenance, and signal regulation. It begins when membrane regions invaginate and pinch off as endocytic vesicles, which deliver cargo to early endosomes for sorting into recycling, retrograde transport, or degradative routes. As cargo moves through maturing endosomes, changes in membrane composition and acidity help direct receptors, lipids, and extracellular material to their destinations, including lysosomes. Understanding this pathway clarifies how cells control surface receptors, internalize pathogens, and process therapeutic nanoparticles, making it central to cell biology, disease research, and drug-delivery studies.

Endocytic and retrograde transport of proteins from the plasma membrane to the trans-Golgi network is essential to maintaining membrane homeostasis and regulating signaling. Here, we describe a method to image and quantify endocytic transport of transmembrane cargo proteins by live-cell microscopy using derivatized anti-GFP nanobodies in HeLa cells.

Regulated endocytosis governs the cell surface expression levels of the majority of membrane proteins. Here we utilize reducible, membrane impermeant biotinylation reagents to measure the endocytic rate of the dopamine transporter (DAT), a polytopic membrane protein. The method facilitates a straightforward approach to measuring the endocytic rate of most plasma membrane proteins.

Several methods are available for measuring intracellular pH but very few of these allow simultaneous measurement of cytoplasmic and organellar pH. Here, we describe in detail a rapid and accurate methodology to simultaneously measure cytoplasmic and vesicular pH by ratiometric imaging of living cells.

The pentose phosphate pathway (PPP) operates in parallel with glycolysis, facilitating the metabolism of both pentoses and glucose. This pathway consists of two distinct phases: the oxidative and non-oxidative phases. While it does not directly generate ATP, the intermediates formed during the process can integrate into glycolysis, contributing to cellular energy metabolism when required.Oxidative Phase: NADPH ProductionThe oxidative phase of the pentose phosphate pathway is primarily...
