Ligand Gated

Ligand-gated ion channels are membrane proteins that open or close when specific chemical messengers, or ligands, bind to them, allowing cells to convert chemical signals into electrical responses. Ligand binding induces a conformational change in the channel, altering ion permeability and driving the movement of charged particles such as sodium, potassium, calcium, or chloride across the membrane. In biology, these channels are central to rapid synaptic transmission, sensory signaling, and communication between excitable cells. Their structure and function also provide important targets for studying neurological disorders and developing drugs that modify receptor activity.

This article describes methods for site-directed spin labeling and reconstitution of pentameric ligand-gated channels for Electron Paramagnetic Resonance studies. This protocol can be adapted for any membrane protein. The reconstitution method described here can also be used for patch-clamp measurements of macroscopic and single-channel currents in a defined lipid system.

Proteomics to Identify Proteins Interacting with P2X2 Ligand-Gated Cation Channels

We describe a simple protocol to identify brain proteins that bind to the full length C terminus of ATP-gated P2X2 receptors. The extension and systematic application of this approach to all P2X receptors is expected to lead to a better understanding of P2X receptor signaling.
