Macrophage Uptake

Macrophage uptake is the process by which macrophages internalize particles, pathogens, cellular debris, and soluble material, supporting tissue maintenance and immune defense. It occurs through receptor-mediated recognition followed by phagocytosis, macropinocytosis, or endocytosis; internalized material enters vesicles that mature and fuse with lysosomes for degradation, processing, or antigen presentation. Studying macrophage uptake helps explain host responses to infection, inflammation, and tissue injury, while uptake assays evaluate how macrophages interact with biomaterials, nanoparticles, and drug-delivery systems. These investigations can clarify immune mechanisms and guide the design of therapies that regulate macrophage activity or target drugs to immune cells.

In this video, we demonstrate the efferocytosis assay to study the clearance of apoptotic cells using confocal microscopy. Efferocytosis is a multi-step process where apoptotic cells are engulfed by phagocytes and degraded within vesicles called efferosomes. This protocol provides a method to visualize internalized and non-internalized portions of individual apoptotic cells and quantify efferocytosis.

The article describes a readily easy adaptive in vitro model to investigate macrophage polarization. In the presence of GM-CSF/M-CSF, hematopoietic stem/progenitor cells from the bone marrow are directed into monocytic differentiation, followed by M1 or M2 stimulation. The activation status can be tracked by changes in cell surface antigens, gene expression and cell signaling pathways.
