Mouse Lung Perfusion

Mouse lung perfusion is an experimental method that circulates a controlled fluid through the blood vessels of an isolated mouse lung to examine pulmonary function without influences from the whole animal. In an ex vivo preparation, researchers ventilate the lung while delivering oxygenated perfusate through the pulmonary artery and collecting it from the venous outflow, allowing measurement of vascular resistance, permeability, and fluid leakage under defined conditions. This approach supports studies of pulmonary circulation, inflammation, edema, respiratory injury, and drug responses. By separating lung-specific effects from systemic factors, mouse lung perfusion provides a controlled platform for investigating mechanisms of disease and evaluating potential therapies.

Ex-Vivo Lung Perfusion (EVLP) has allowed lung transplantation in humans to become more readily available by enabling the ability to assess organs and expand the donor pool. Here, we describe the development of a rat EVLP program and refinements that allow for a reproducible model for future expansion.

Here, we describe the necessary steps for establishing a rat EVLP model and show the inflammatory profile associated with the perfused lungs. The aim is to propagate knowledge and experiences about the rat EVLP model, enabling the integral understanding of the biological responses associated with that revolutionary technique.

Antegrade Perfusion Model: An Ex Vivo Technique to Perfuse Isolated Mouse Heart

This video describes an antegrade perfusion technique of an isolated mouse heart during which a perfusate is injected through the left ventricle, and eventually, it perfuses the entire myocardium. This method can be used to isolate cardiomyocytes from murine models and generate decellularized hearts for further downstream applications.
