Protein Metabolism

Protein metabolism encompasses the biological processes that synthesize, modify, transport, and break down proteins and amino acids, enabling cells to maintain structure, function, and energy balance. Cells regulate these processes through gene expression and translation for protein production, while enzymes control proteolysis and amino acid catabolism when proteins are damaged, no longer needed, or required as metabolic fuel. In biology, studying protein metabolism clarifies growth, tissue repair, nitrogen balance, and responses to nutritional or physiological stress. Its disruption can contribute to metabolic disorders and disease, making the topic important for research in nutrition, medicine, and cellular regulation.

Overview of Protein Metabolism

2024

Proteins are broken down into amino acids during digestion. Unlike fats and carbohydrates, which are stored for later use, proteins are not. Instead, amino acids are either used to produce ATP through oxidation or contribute to the creation of new proteins for the growth and repair of the body. Any surplus amino acids from the diet are converted into glucose or triglycerides rather than excreted. Amino acids play various roles in the body once they are absorbed into cells. They are restructured...

Metabolic Labeling

2023

Metabolic labeling is used to probe the biochemical transformations and modifications that occur in a cell. This is accomplished by using chemical analogs that mimic the structure of natural biomolecules. Cells utilize analogs in their endogenous biochemical processes, producing compounds that are labeled. The label allows for the incorporation of detection and affinity tags, which can then be used to elucidate metabolic pathways using other biochemical analytical techniques, such as SDS-PAGE...

Zebrafish represent a powerful vertebrate model that has been under-utilised for metabolic studies. Here we describe a rapid way to measure the in vivo metabolic profile of developing zebrafish that allows the comparison of different mitochondrial function parameters between genetically or pharmacologically manipulated embryos, thereby increasing the applicability of this organism.

An Introduction to Cell Metabolism

2023

In cells, critical molecules are either built by joining together individual units like amino acids or nucleotides, or broken down into smaller components. Respectively, the reactions responsible for this are referred to as anabolic and catabolic. These reactions require or produce energy typically in the form of a “high-energy” molecule called ATP. Together, these processes make up “Cell Metabolism,” and are hallmarks of healthy, living cells.JoVE’s introduction to cell metabolism briefly...

We present a variation of the QUICK (QUantitative Immunoprecipitation Combined with Knockdown) approach that was introduced previously to distinguish between true and false protein-protein interactions. Our approach is based on 15N metabolic labeling, the modulation of affinities of protein-protein interactions by the presence/absence of ATP, immunoprecipitation, and quantitative mass spectrometry.
