Sgc Alpha One Knockout

Sgc Alpha One Knockout is a genetic model in which the gene encoding the alpha 1 subunit of soluble guanylate cyclase is disrupted, allowing researchers to study its biological function. Because this subunit normally partners with the beta 1 subunit to form a nitric oxide-responsive enzyme, its loss reduces alpha 1-containing guanylate cyclase activity and alters production of the signaling molecule cyclic guanosine monophosphate (cGMP). This model helps clarify nitric oxide signaling in vascular, neural, and other physiological processes, while distinguishing alpha 1-dependent effects from signaling mediated by alternative guanylate cyclase isoforms. It is useful for investigating disease mechanisms and evaluating pathway-targeted therapies.

Immunostaining of Neurons Treated with Alpha-Synuclein Aggregates

2025

This video demonstrates the immunostaining of neurons treated with alpha-synuclein (αS) aggregates. The neurons are first treated with αS aggregates. They are then fixed, permeabilized, and treated with a blocking solution. Primary antibodies are added, followed by a fluorescently labeled secondary antibodies and a nuclear stain. The sample is then mounted and observed under a microscope.

CRISPR Concatemer-Mediated Multiple Gene Knockout: A Technique to Simultaneously Knockout Multiple Genes by Non-Homologous End-Joining Pathway in Mouse Intestinal Cells

Induction of Neuroinflammation in Mice via Injection of Alpha-Synuclein Fibrils

2025

This video demonstrates the injection of alpha-synuclein fibrils into a mouse to induce neuroinflammation. The fibrils are injected intraperitoneally into a transgenic mouse expressing mutated alpha-synuclein proteins. Upon reaching the central nervous system, the fibrils enter neurons and induce aggregation of the mutated proteins. The aggregates spread to neighboring neurons, microglia, and astrocytes. Recognition of the aggregates triggers microglia to induce neuroinflammation, damaging the...

A Protocol for Multiple Gene Knockout in Mouse Small Intestinal Organoids Using a CRISPR-concatemer

Cited by 18 •

2017

This protocol describes the steps for cloning multiple single guide RNAs into one guide RNA concatemer vector, which is of particular use in creating multi-gene knockouts using CRISPR/Cas9 technology. The generation of double knockouts in intestinal organoids is shown as a possible application of this method.
