In Vivo Mouse Model

An in vivo mouse model is a living mouse used to study biological processes, disease mechanisms, or responses to experimental interventions within an intact organism. Researchers introduce or observe a defined genetic, physiological, or pathological condition and then measure outcomes across interacting organs, tissues, and cell types, preserving biological relationships that isolated cells or tissues cannot fully reproduce. In biology, these models support investigations of disease progression, gene function, drug activity, toxicity, and therapeutic efficacy. Findings can reveal system-level effects and guide the development of hypotheses and treatments, although differences between mouse and human biology require careful interpretation.

Orthotopic animal models of ovarian cancer replicate better human disease and therefore enhance our understanding of cancer progression and tumor response to therapy. A mouse model receives an intrabursal injection of luciferase-expressing ovarian tumor cells. Treatment is administered via oral gavage. Tumor growth is monitored by in vivo imaging system.

In Vivo Fluorescence Imaging to Localize Antibodies in a Mouse Tumor Xenograft Model

2025

The video demonstrates a fluorescence imaging method for in vivo antibody localization in a mouse tumor xenograft model. It involves the injection of tumor cells in a mouse to generate solid xenografts beneath the skin. Subsequent injection of fluorescent antibodies confirms specific localization through enhanced fluorescence signals within the tumor xenograft.

Here, we present a systematic approach for developing physiologically relevant, sensitive and specific in vivo assays for interpreting variation in human pathology. Transient genetic manipulation via microinjection of WT and mutant human mRNA and morpholino (MO) antisense oligonucleotides harness the tractability of the developing zebrafish embryo to rapidly assay pathogenic mutations, especially, but not exclusively, in the context of human developmental disorders.
