Denaturant Concentration

Denaturant concentration is the amount of a chemical unfolding agent, such as urea or guanidinium hydrochloride, present in a solution, and it is a key variable for controlling protein structure and stability. As concentration increases, these agents weaken the noncovalent interactions that maintain native folding, including hydrophobic interactions and hydrogen bonding, shifting the equilibrium toward unfolded conformations. In chemistry and biochemistry, controlled denaturant gradients help characterize protein stability, solubilize or unfold samples, and optimize purification and refolding procedures. Measuring concentration precisely improves reproducibility when comparing folding behavior or testing how sequence and conditions affect molecular structure.

Denaturing urea polyacrylamide gel electrophoresis is used to separate single-stranded DNA or RNA up to a limit of 500 nucleotides. Urea in combination with heat denatures samples and unstructured single strands migrate within the gel matrix according to their molecular weight.
